In the context of current demographics, with the percentage of the population age 60+ increasing and set to reach 2 billion people by 2050, the concept of drug drug interactions has become a hot topic. The increase in life expectancy comes with some challenges, as for example polymedication for chronic diseases. With up to 40% of over 65 year olds currently taking more than 4 prescription drugs, the risks of drug drug interactions are a cause for concern.

First of all, we would like to wish you a Happy New Year 2014 to all the readers of Intelligent Pharma’s blog (you were, on average, 415 unique readers per day during 2013). 2014 is starting and it has to be the year of many challenges. In our field, the computational chemistry, our point of view is that the main challenge for 2014 has to be the polypharmacology. During the past years (2011-2013) many publications appeared about this topic discussing basic science aspects and showing the potential of these approaches.

Polypharmacology is a newly emerging approach which reflects the high complexity of the mechanism of actions of drugs. In fact, some months ago we already posted an article about polypharmacology in our blog. The overall interest on this discipline has risen significantly since then and the number of articles and publications on the topic are by now overwhelming. The reasons of such increasing interest are multiple –drug reprofiling, in silico toxicology, etc.– but, there is one reason that is especially important and it is, probably, one of the key factors of the increasing interest on polypharmacoloty. This reason is phenotypic screening.
